Studentka bude v rámci diplomové práce klonovat, produkovat a purifikovat různé bakteriální dinukleotid cyklasy. Následně bude charakterizovat jejich substrátovou specifitu za použití nepřirozených nukleotid trifosfátů (NTPs) a enzymaticky připraví malá množství cyklických dinukleotidů. Nakonec bude charakterizovat biologickou aktivitu připravených cyklických dinukleotidů v buněčných a biochemických testech.

In this diploma thesis student will focus on cloning, expression and purification of several bacterial dinucleotide cyclases. Substrate specificity of purified enzymes will be characterized using analogues of nucleotide triphosphates (NTPs) and cyclic dinucleotides (CDNs) will be then prepared enzymatically. Student will also determine biological activity of prepared CDNs in biochemical and cell-based assays.
